In the modern textile printing and dyeing industry, cellulase serves as a core auxiliary agent for green biological processing. It is widely applied in key processes such as fabric bio-polishing and denim abrasion. Enzyme activity is a crucial indicator for evaluating its catalytic performance and controlling production processes and product quality, possessing significant engineering application value and quality control implications. Enzyme activity determines the fabric treatment effect. Sufficient enzyme activity can effectively removes surface fuzz, inhibits pilling and improves textile quality. Low enzyme activity results in poor fabric treatment, while high activity can damage fiber strength and lead to significant fabric weight loss. Stable and controllable enzyme activity is a vital quality control basis for standardized and refined production processes. Therefore, cellulase activity detection has become a focus.
The light industry standard QB/T 2583-2023 of China specifies the detection methods for cellulase activity, including the filter paper method and the sodium carboxymethyl cellulose (CMC-Na) method. Baico Biotech BBT Enzyme is a textile enzyme and household detergent enzyme manufacturer in China. BBT Enzyme adopts the CMC-Na method. Although BBT has its own specific testing procedures, the experimental principle is the same. Taking advantages of enzyme specificity, the assay conducts enzymatic catalytic reactions with CMC-Na as the substrate.
Natural cellulose is insoluble in water. CMC-Na, a cellulose derivative, has been modified to introduce hydrophilic groups, completely overcoming its water solubility deficiency. It can be formulated into a homogeneous, stable, and viscous aqueous solution, providing a favorable reaction environment for enzyme catalysis. Simultaneously, CMC-Na retains the core molecular structure of cellulose, enabling it to be specifically recognized and hydrolyzed by cellulose to generate reducing sugars.
With strict control over all experimental variables including temperature, pH, substrate concentration, enzyme dosage and reaction duration, reducing sugar concentration directly reflects the catalytic capacity of cellulase. Higher content corresponds to higher cellulase activity. This lays the fundamental theoretical foundation for subsequent quantitative detection of reducing sugars and calculation of enzyme activity.
